Neste trabalho foram pesquisados 2 microrganismos,
Staphylococcus aureus e Escherichia coli, no campus
do Instituto Politécnico de Macau em ambientes
e superfícies de locais com elevada frequência
populacional pela colheita de 62 amostras, em 9
locais distintos, pelos métodos de amostragem do
ar, por sedimentação, e de superfícies, pelo método
da zaragatoa, com inclusão nos meios Mannitol Salt
Agar (MSA) e Agar Eosin-Methylene Blue (EMB),
respetivamente e incubação até 48h a 37ºC. Nas
19 placas de MSA com crescimento característico,
para além da contagem do número de Unidade
Formadoras de Colónias, foi realizada coloração de
Gram e os testes bioquímicos Catalase e Coagulase
para identificação como S. aureus. A sua presença foi
identificada em 88,89% (8/9) dos locais de recolha
de amostras, com 70,37% (19/27) de crescimento
nas amostras recolhidas. A ausência de crescimento
nas placas de EMB pressupõe a ausência de E. coli
nos locais em que foi realizada a pesquisa e de forma
abrangente em todo o campus.
ABSTRACT Staphylococcus aureus and Escherichia coli were screening on the campus of the Polytechnic Institute of Macao in environments and surfaces of high population frequency sites by harvesting 62 samples at 9 different sites using the methods of air sampling by sedimentation and surfaces by swab, inclusion in Mannitol Salt Agar (MSA) and Eosin-Methylene Blue (EMB) mediums, respectively, and incubated for 48h at 37 °C. In the 19 MSA plates with characteristic growth, in addition to counting the number of Colony Forming Units, Gram staining and the biochemical tests Catalase and Coagulase were performed for identification as S. aureus. The presence of these microorganism was identified in 89.90% (8/9) of the sampling sites with 70.37% growth in the samples collected. The lack of growth in EMBs assumes the absence of E. coli at the sites where the research was carried out and in a big scale, across the campus.
